An RT-qPCR based multiplex amplification test allowing specific detection of the SARS-CoV-2 genomic RNA.

INTENDED USE: MAScIR SARS-CoV-2 M 2.0 KIT is an in-vitro qualitative multiplex test allowing the detection of the coronavirus 2 nucleic acid using Nasopharyngeal as well as an oropharyngeal swab.

PROCEDURE:

RNA extraction

• MAScIR SARS-CoV-2 M 2.0 KIT is used only with RNA containing
elution following extraction from a given sample. it is mandatory to
have an RNA extraction kit to perform the RNA extraction before the
use of the MAScIR SARS-CoV-2 M 2.0 KIT for the amplification step.
• MAScIR SARS-CoV-2 M 2.0 KIT could be used with Mini Kit NucleoSpin
Dx Virus (Macherey Nagel), Mini kit QIAamp Viral RNA (Qiagen), Kit
Biospin Virus DNA/RNA Extraction (Bioflux)

Test procedure
For each test, you can prepare a reaction mix containing all the required
reagents as described in the following tables (for serval samples,
prepare a solution by mixing Mix1+Mix2 enough to test all the analyzed
samples):

REQUIRED EQUIPMENT AND CONSUMABLES:
• Adjustable Micropipettes and filtered tips.
• Bench Centrifuge.
• Vortex.
•Gloves.
• Sterile Micro tubes.
• 96-well Microplates with adhesive film.
• RT-qPCR machine.

HANDLING CONDITIONS:

• MAScIR SARS-CoV-2 M 2.0 KIT has to be used only by professional
laboratory labor who are well trained in molecular biology-based analysis.
• Read the instructions provided in the package carefully before processing
samples
• Avoid contact with skin and eyes. in case of any contact wash
extensively with water.
• Use aerosol-resistant pipette tips and use a new tip every time a volume is
dispensed.
• Do not eat, drink, smoke, or apply cosmetics in areas where reagents of
samples are handled.
• Use sterile disposable laboratory materials and do not re-use the tubes and
tips.
• Do not mix reagent from different batches of the kit.
• Do not use a kit after its expiration date.
• Homogenize the solution by mixing very well using vortex before each
use.
• Collect the solution by centrifuging briefly before use.
• Use a correct PCR microplate.
• centrifuge the microplate before using it in the PCR machine.
• Avoid bubbles forming by harsh pipetting.
• In case of several use, we recommend that you make aliquots of both
mix1 and 2. Freezing and defrosting could alter the quality of the
reagents contained in the kit.
